How to mix a AOD-9604 vial

These steps are for the 2 mg vial picked in the chart. Change the size there and the amounts here change with it.

  1. Wash your hands. Wipe the stopper of the peptide vial and the water vial with an alcohol swab.
  2. Draw 2 mL of bacteriostatic water into a syringe.
  3. Push the needle through the stopper of the peptide vial and let the water run slowly down the glass, not straight onto the powder.
  4. Swirl the vial gently until the powder is gone. Do not shake it.
  5. Write the date and 1 mg/mL on the vial, then put it in the fridge.

What AOD-9604 is

AOD-9604 is a synthetic peptide: the tail end of human growth hormone (amino acids 177 to 191) with one extra tyrosine added at the front. It was tested in Australia as an obesity drug taken by mouth. No approved product was found. [5]

Other names: AOD9604, Tyr-hGH 177-191.

How it has been given to people: by mouth (capsules or tablets) in the multi-dose human trials; two early studies gave a single intravenous infusion. No published human trial injected it under the skin.

What to know before reading a dose

  • Not approved in the US or Canada, and no approved product was found anywhere.
  • The human trials gave it by mouth. No published human trial of injecting it under the skin was found, so injectable doses in circulation have no trial behind them.
  • The company that ran the trials wrote in 2014 that the weight loss seen in early trials was not seen in the last and largest trial [2]. Claims that it is proven to burn fat in people are not supported by anything found here.
  • Both human-data papers come from the compound's owner and its consultants and sit in a journal PubMed does not index [1], [2].
  • GRAS: the 2013 paper says an expert panel judged it Generally Recognized As Safe for use in foods. That is a panel finding reported by the company. FDA's GRAS notice inventory has no record for AOD9604 [13].
  • FDA placed it in category 2 for safety risk, the nomination was later withdrawn, and FDA says it lacks enough information to know whether it would cause harm [7].
  • WADA-listed: named on the Prohibited List under growth hormone fragments [12].

What the evidence shows

Randomized human trial

Weight loss in people: the company that ran the trials wrote that the effect seen in early trials was not seen in the last and largest one. No paper reporting the weight results themselves was found. [2]

Side effects and warnings

Reported in human studies or on a label:

  • Across the six trials the authors report no withdrawal and no serious adverse event that was put down to AOD9604. [1]
  • In the 24-week trial of 0.25 mg to 1 mg a day by mouth, the authors report a safety profile no different from placebo. [1]
  • At 54 mg a day by mouth for 7 days, headache, diarrhea and gas were more frequent. [1]
  • In the intravenous study in obese subjects, 5 of 23 reported mild or moderate euphoria while on AOD9604 and none while on placebo. [1]
  • FDA says it has identified serious adverse events that may be linked to AOD-9604, with causality unclear. [7]
  • FDA says compounded AOD-9604 may carry a significant risk of an immune reaction by some routes of administration. [7]

AOD-9604 has no approved label in the US or Canada for this use, so there is no official list of who should not use it, no interaction list and no missed-dose rule.

Storage

Keep an unmixed vial the way its label says. Once it is mixed, keep it in the fridge, do not freeze it, and write the mixing date on the vial.

Looked for and not found

  • Weight-loss results from the human trials. PubMed holds no trial report, and the two company papers that were found give safety and animal data only.
  • Any human study of injection under the skin. The six trials used intravenous infusion (two single-dose studies) or dosing by mouth.
  • A human half-life. The only figure found is about 3 minutes after an intravenous dose in pigs [2].
  • Any trial registration on ClinicalTrials.gov [14]. The 2013 paper says the two largest trials were filed under Australia's Clinical Trial Notification scheme; that record was not looked up.
  • No approved label anywhere, so no official contraindications, interaction list, missed-dose rule or storage instructions.
  • The doses given by vein were set per kg of body weight and are not in the dose table. They are in [1].
