There is no established dose of MGF. No published human study has given the peptide at any dose, and the evidence is cell and animal work that one industry lab could not repeat. Reports describe 100 to 200 mcg after training, which is not a label dose. It is not approved in the US or Canada.

How to mix a MGF vial

These steps are for the 2 mg vial picked in the chart. Change the size there and the amounts here change with it.

  1. Wash your hands. Wipe the stopper of the peptide vial and the water vial with an alcohol swab.
  2. Draw 2 mL of bacteriostatic water into a syringe.
  3. Push the needle through the stopper of the peptide vial and let the water run slowly down the glass, not straight onto the powder.
  4. Swirl the vial gently until the powder is gone. Do not shake it.
  5. Write the date and 1 mg/mL on the vial, then put it in the fridge.

What MGF is

MGF (mechano growth factor) is the name given to IGF-1Ec, a form of insulin-like growth factor 1 that muscle makes after loading. The peptide sold as MGF is a synthetic copy of the 24 amino acids at the end of its E domain. [8]

Other names: mechano growth factor, mechano-growth factor, IGF-1Ec, IGF-I Ec, MGF E peptide, MGF-E, MGF-Ct24E.

How it has been given to people: no human use on record.

What to know before reading a dose

  • No human study of injected MGF was found. Every dose in circulation comes from outside the published literature.
  • The muscle claims rest on cell culture. A 2014 attempt by two pharmaceutical companies to repeat them failed, and its authors questioned whether the peptide does anything in the body [3].
  • The human studies that mention MGF measured the body's own MGF messenger RNA after exercise. They are not evidence that injecting the peptide builds muscle [6].
  • The animal work is on stroke in gerbils and heart attack in sheep, not on muscle growth [4], [5].
  • PEG-MGF is a different molecule: MGF with a polyethylene glycol chain attached. Its doses do not transfer to plain MGF.
  • Not an approved medicine in the US or Canada as of the check date.
  • WADA Prohibited List 2026, section [2].3, names mechano growth factors (MGFs); the class is prohibited at all times [12]. A 2014 paper says the substance has been treated as prohibited since 2005 [8].

What the evidence shows

Each finding is tagged by the kind of study it comes from. Human trials are listed first.

Human observational study

The human data are about the body's own MGF, not the injected peptide: after heavy leg exercise, MGF messenger RNA rose in the muscle of 8 young men but not of 7 elderly men. Nobody was given MGF. [6]

Animal study

In gerbils with a temporary cut in brain blood flow, the synthetic peptide protected vulnerable neurons. [4]

In sheep with an induced heart attack, the E domain peptide preserved heart function over eight days. [5]

Lab study

The founding cell study: in a mouse muscle cell line, the E domain peptide raised myoblast proliferation and held back the final step of differentiation. [1]

In cultured human muscle cells the 24 amino acid peptide extended the dividing life of satellite cells from newborn and young adult muscle, but not from old adult muscle. [2]

Side effects and warnings

Reported in human studies or on a label:

  • No human study of MGF peptide was found, so no side effects have been recorded in people. Writing about the pegylated form, FDA says it has no human exposure data and names immune reactions and peptide impurities as possible risks of compounded product. [9]

MGF has no approved label in the US or Canada for this use, so there is no official list of who should not use it, no interaction list and no missed-dose rule.

Storage

Keep an unmixed vial the way its label says. Once it is mixed, keep it in the fridge, do not freeze it, and write the mixing date on the vial.

MGF dosage questions

How many units is 200 mcg of MGF from a 2 mg vial?

Mixed with 2 mL of bacteriostatic water, a 2 mg vial is 1 mg/mL. 200 mcg is 0.20 mL, which is 20 units on a U-100 syringe.

How much water goes in a 2 mg MGF vial?

2 mL gives 1 mg/mL. At that strength the doses in the chart sit at 10, 20 units on a U-100 syringe.

How many doses are in a 2 mg vial?

Whole doses in one vial: 20 at 100 mcg, 10 at 200 mcg.

Is MGF approved in the US?

Not FDA approved. FDA lists the pegylated form, PEG-MGF, among bulk substances that were in compounding category 2 and whose nominations were then withdrawn; the plain peptide is not named.

Is MGF authorized in Canada?

No MGF product, current or cancelled, is in Health Canada's Drug Product Database. Injectable peptides are regulated as prescription drugs in Canada.

What human dosing data is there for MGF?

No human study of injected MGF was found. Every dose in circulation comes from outside the published literature.

Looked for and not found

  • No published human study in which MGF peptide was given, by any route or at any dose. PubMed searches for mechano growth factor with human, injection, pharmacokinetics and half-life returned cell, animal and doping-test papers only.
  • No trial registered on ClinicalTrials.gov. A search for mechano growth factor as the intervention returns 40 studies through the IGF-1 synonym, all of them growth hormone, IGF-1 or unrelated work; none tests MGF peptide.
  • No human half-life or other pharmacokinetic figure for the peptide.
  • No human safety data.
  • No approved label in the US or Canadian sources searched, so no official dose, contraindications or storage instructions.
  • No published human data on the pegylated form, PEG-MGF, either. FDA says it has identified none [9].
