Retatrutide has been given once a week by injection under the skin. The Phase 3 obesity trial used 4 mg, 9 mg or 12 mg a week, and the earlier Phase 2 trial used 1 mg to 12 mg, starting at 2 mg or 4 mg. It is not an approved medicine, so there is no label dose.

Tirzepatide is injected under the skin once a week. The US and Canadian labels start at 2.5 mg for 4 weeks, then 5 mg, and allow steps of 2.5 mg no sooner than every 4 weeks, up to a maximum of 15 mg a week. These are the label doses for Mounjaro and Zepbound.

Semaglutide for weight management is injected under the skin once a week. The Wegovy label starts at 0.25 mg and steps up every 4 weeks through 0.5 mg, 1 mg and 1.7 mg to 2.4 mg. The US label now allows up to 7.2 mg a week. Canada's stops at 2.4 mg.

Cagrilintide has been given once a week by injection under the skin in trials. The Phase 2 trial tested 0.3 mg to 4.5 mg a week and the Phase 3 trials used 2.4 mg. It is not approved in the US or Canada, so there is no label dose.
